The top two in Group D clash in a potential top-of-the-table decider as FC Porto host Chelsea at the Estadio do Dragao, whilst in Cyprus, APOEL Nicosia welcome Atletico Madrid to the GSP Stadium.
Chelsea currently lead the way in Group D with 10 points from four matches whilst FC Porto lie second, just one point back, on nine.
The pair secured their safe passage into the knockout stage on Matchday 4 as the Blues drew 2-2 away to Atletico Madrid, and the 2004 Champions League winners sealed a narrow 1-0 win over APOEL courtesy of Falcao's late strike.
Now with the chance to cement top-spot and thus avoid the big guns in the last 16, Wednesday evening's clash at the Estadio do Dragao promises to be an intriguing affair.
Nicolas Anelka’s second-half strike was enough to give Chelsea a slender 1-0 win over the Portuguese champions on Matchday 1, however, with a impending clash with Arsenal to come on Sunday, coach Carlo Ancelotti may be tempted to rest a few of his first-team stars as he seeks to juggle European and domestic commitments.
Porto themselves lost again at the weekend, this time away to Maritimo, but that has not perturbed coach Jesualdo Ferreira who is targeting victory and top-spot.
"The idea is to get the most points in qualification and go forward. Porto, in the last few years, has always made its presence felt in the knock-out phase of the Champions League," he was quoted as saying by Sky Sports.
"We want to play at a more superior level than we have been during the last few weeks.
"We had a very good performance at Stamford Bridge so we must now look to win the game here and to compete at the most superior level we can," he added.
Elsewhere in the group, Cypriot champions APOEL Nicosia host Atletico Madrid at the GSP Stadium with both sides hoping to bag their first win in this season's UEFA Champions League, while the loser will take home the wooden spoon.
